异丙基肾上腺素诱导心力衰竭大鼠心肌一氧化氮的改变

摘    要:目的 建立异丙基肾上腺素诱导SD大鼠充血性心力衰竭模型,观察心肌一氧化氮的变化.方法 选用清洁级雄性SD大鼠50只,随机分为空白组10只,皮下注射生理盐水2次(0.25 ml,间隔24 h);模型组40只,皮下注射ISO 2次(170mg/kg间隔24 h).6周后应用超声心动图检测,以左室射血分数≤45%确定造模成功,将模型随机分为心衰代偿组8只[生理盐水0.1 ml/(kg·d)ig]、心衰失代偿组14只[生理盐水0.1 ml/(ks·d)ig];18周使用超声心动图检测心功能指标,包括短轴缩短率(FS)、左室射血分数(LVEF)、左室舒张末内径(LVEDD)、左室收缩末内径(LVESD);取心肌称量心室质量指数(VWI)、观察病理形态学特征,并利用硝酸还原酶法检测心肌NO浓度.结果 与空白组相比,心衰代偿组和心衰失代偿组心功能指标明显下降,心肌NO、心室质量指数明显升高(心衰代偿组P<0.05,心衰是代偿组P<0.01).与心衰代偿组相比心衰失代偿组心功能指标显著恶化,心肌NO、心室质量指数明显升高(P<0.01).结论 应用大剂量ISO可诱导大鼠充血性心力衰竭,该作用可能与心肌NO过度升高有关.
