| 摘 要： | 目的探索人肌腱胶原蛋白在组织工程皮肤及整形外科中的应用。方法从人肌腱提取酸溶性胶原,制备人肌腱胶原蛋白。结果所提取的胶原在pH5.8左右迅速形成凝胶。光镜下可见酸溶性胶原纤维呈长细丝状,交织成网状。纯度以羟脯氨酸质量分数为基准,分光光度法测定为95%。氨基酸成分主要为甘氨酸占35%,脯氨酸占10%。结论氨基酸分析表明,所提取的胶原为典型的Ⅰ型胶原蛋白,热稳定性测定Td为39～41℃。

| Abstract: | Objective To investigate the use of human tendon collagen in tissue engineered skin and plastic surgery.Method A new technology for extraction of acid-resolved collagen from human tendon was studied in this paper,ie, 0.1%HAc solution was used as solvent to refine human tendon collagen.Results The extracted collagen can be able to form collagen gel immediately at pH5.8.The morphology of the collage gel under a light microscope was follows:the acetic acid-resolved collagen fibrils were long, straight and thin .The purity of collagen was 95% based on the hydroxyproline content determined by UV. The component of amino acid was mainly aminoacetic acid reaching 35%,proline reaching 10%. Conclusion From amino acid analysis,it is concluded that the extracted collagen is typical I type collagen.Determination of thermal stability of collagen expressed as Td is 39-41℃. |
