摘    要:目的建立反相高效液相色谱快速测定血清中氨甲蝶呤浓度的方法,并用于临床患者血药浓度监测。方法以C18为固定相,乙腈-40mM醋酸缓冲液(pH4.6)=11.1:88.9(v/v)为流动相,检测波长294nm。结果 MTX在0.04~1.6μg.mL-1范围内呈良好的线性关系,其回归方程为Y=3.2758X-0.0057,r=0.9999;日内精密度分别为4.59%、3.80%、3.67%;日间精密度分别为5.84%、2.48%、1.67%;加样回收率为101.20%。结论该方法简便,快速,灵敏度高,重复性好,可用于临床MTX血药浓度监测,保障临床大剂量MTX用药的安全性和有效性。

Abstract:OBJECTVE To develop a rapid method for the determination of MTX concentration in plasma by RP-HPLC.METHODS C18 column was used with acetonitrile-40mM acetate buffer(pH4.6)(11.1∶88.9)as mobile phase.The UV detection wavelength was 294 nm.RESULTS The calibration curve was linear within the range of 0.04~1.6μg·mL-1;The linear equation was Y=3.2758X-0.0057,r=0.9999;The intra-day RSD was 4.59%、3.80%、3.67% and The inter-day RSD was 5.84%、2.48%、1.67%;The recovery rate was 101.20%.CONCLUSION It is applicable to determinate MTX concentration in plasma for clinical practice.
