紫杉醇对裸鼠人肝癌血管生成和肿瘤生长、转移的作用

摘    要:目的:通过建立高转移潜能人肝癌裸鼠肝原位移植模型,观察紫杉醇对人肝细胞癌(HCC)血管生成和肿瘤生长、转移的干预作用。方法:24只人肝癌裸鼠模型随机分成两组,实验组和对照组,每组12只。自模型建立次日始两组分别给予13mg·kg-1·d-1剂量紫杉醇和等体积生理盐水腹腔注射,给药第30d处死裸小鼠,观察肿瘤生长、转移情况,分别用免疫组化和RTPCR的方法检测癌组织CD34和VEGFmRNA的表达水平,记录微血管密度(MVD)。结果:紫杉醇组肿瘤重量、体积、肺转移灶计数、MVD和VEGF均低于对照组(P<0.05)。结论:紫杉醇对HCC生长有抑制作用,抑制肝细胞癌的血管生成并降低肺转移。

Abstract:Objective: Effects of paclitaxel on the angiogenesis and tumor growth and metastasis of hepatocellular was observed in nude mice model bearing huaman hepatocellular carcinoma. Methods: 24 nude mice were randomly divided into two groups, the therapy group and control group, with 12 mice in each. paclitaxel (13 mg·kg~(-1)·d~(-1) was administered intraperitoneally in the therapy group and equivalent 0.9% natrichloride solution was used in the control group. Mice were sacrificed on the 30th day, and tumor size and weight and metastases in liver and lungs were measured. Levels of CD34 and VEGF mRNA in tumor tissue were detected by immunohistochemical and semi-quantitative technology respectively, and microvessel density (MVD) was also counted. Results: MVD and VEGF mRNA in therapy group were less than in the control group (P<0.05), metastases in lung,tumor size and weight in the therapy group were also less than that in the control group. Conclusion: Paclitaxel can significantly inhibit angiogenesis and metastasis of hepatocellular both with its tumor growth inhibitory effect.
