HIF—1α、VEGF在胰腺癌组织中的表达及临床意义

目的研究胰腺腺癌组织中的低氧诱导因子-1α（hypoxia—inducible factor—1α，HIF-1α）和血管内皮生长因子vascular endothelial growth factor，VEGF）的表达情况及其临床意义。方法采用Westernblotting方法检测30例胰腺腺癌组织及其中9例胰腺癌相应癌旁胰腺组织中HIF—1α、VEGF的表达。结合临床病例资料，分析HIF—1α、VEGF与肿瘤的大小、部位、病理分化程度、淋巴结转移及胰腺癌TNM分期之间的关系。结果统计结果显示HIF-1α、VEGF在胰腺癌肿瘤组织中的表达高于癌旁组织，两者比较具有统计学意义（P〈0．01），同时相关性分析显示肿瘤组织中HIF—1α与VEGF两者之间具有正相关性（r=0．655，P〈0．01）。结合临床资料分析，胰腺癌肿瘤组织中HIF—1α、VEGF的表达与肿瘤大小、淋巴结转移和胰腺癌TNM分期密切相关，并且具有统计学意义（P〈0．01）。结论胰腺癌组织中HIF—1α、VEGF存在高表达，HIF—1α、VEGF和肿瘤大小、淋巴结转移和胰腺癌TNM分期有关。HIF-1α、VEGF可能可作为判断胰腺癌预后的一个指标。

Expressions of HIF-1α and VEGF in pancreatic cancer and their clinical significance

Objective To investigate the expressions of hypoxia-inducible factor-1 alpha （HIF-1α） and vascular endothelial growth factor （VEGF） in pancreatic cancer and clinical significance. Methods The expressions of HIF-lot and VEGF in 30 cases of pancreatic cancer and corresponding pancreatic para-cancer tissues （9 cases） were studied with Western blotting method. The relationship and clinical significance of HIF- 1α and VEGF expression in pancreatic cancer were analyzed with the clinical data. Results Expressions of HIF-1α and VEGF in pancreatic cancer tissue were significantly higher than that in para-cancer tissue （P〈 0.01）, and the correlative analysis revealed that the expression of HIF-1α was positively correlated with VEGF （r=0.655, P〈0.01）. The levels of HIF-1α and VEGF were closely related with the size of tumor, the lymphnode metastasis and TNM stage. Conclusion Over-expressions of HIF-1α and VEGF were found in pancreatic cancer. HIF-1α and YEGF may play important roles in the carcinogenesis and aggression in pancreatic cancer. HIF-1α and VEGF may be a useful marker for evaluating prognosis in pancreatic cancer.
