| 摘 要： | 目的:探讨获得胚胎干细胞(ES)简单有效的方法.方法:通过普通离心机以600 r/min离心6 min摔裂小鼠完整囊胚,剥离出未分化的内细胞团,培养鉴定ES.结果:低速离心可将内细胞团从完整囊胚中剥离出来,接种在丝裂霉素处理过的饲养细胞上培养时,能在2～3 d获得巢状增殖的干细胞集落,且能反复传代、冻存复苏,并保持胚胎干细胞多个标志物的特征.结论:利用低速离心摔裂完整囊胚法可获得胚胎干细胞系,且操作方便、经济可行.

| Abstract: | Aim:To investigate a simple and effective method to get embryonic stem cells.Methods:Mouse intact blastocysts was broken by low-speed centrifugation,undifferentiated inner cell masses were isolated and cultured,and embryonic cells markers expression was detected.Results:The inner cells masses could be isolated at 600 r/min speed in general centrifuge,inoculation and culture at trophoblast treated by mytomycin,nested proliferative stem cells colonies were got at 2~3 d.The number of cell passages were demonst... |
