基因表达谱芯片在白血病耐药相关基因研究中的应用

摘    要:目的探讨基因表达谱芯片技术在白血病耐药相关基因研究中的应用.方法首先采用荧光定量基因扩增技术检测了72例白血病患者的MDR1基因的表达,并利用含有4096条人类全长基因的cDNA表达谱芯片对其中3例MDR1基因高表达并表现为临床高度耐药的耐药实验组病例及3例MDR1基因低表达且未表现临床耐药的非耐药对照组病例的白血病耐药相关基因进行了研究. 结果基因表达谱分析两次重复实验结果显示总共有150条靶基因(约3.66%)在与以耐药组及非耐药组细胞RNA为模板合成的双探针杂交时表现出较大的差异.结论运用基因芯片对白血病耐药基因表达谱进行分析,能够筛选出白血病耐药相关基因.

Abstract:Objective: To evaluate the role of gene expression microarray analysis in the study on multidrug resistance in leukemia. Methods: MDR1 gene in 72 patients with leukemia was detected by fluorescence-quantitative RT-PCR technique, and an analysis of MDR gene expression in 3 cases with clinical multidrug resistance and 3 cases with non-clinical multidrug resistance were done by cDNA microarray which represented a set of 4096 human gene. Results: Comparing multidrug resistance group with non-clinical multidrug resistance group, 150 differentially exressed genes were identified by cDNA microarray. Conclusion: cDNA microarray for analysis of gene expression is effective to study multidrug resistance in leukemia.
