PTEN和COX-2在前列腺癌中的表达及其与肿瘤血管生成的关系

摘    要:目的 探讨PTEN和环氧化酶-2(COX-2)在前列腺癌(PCa)中的表达及其与肿瘤血管生成的关系.方法 应用免疫组化法在自制组织芯片上检测60例PCa(PCa组)及25例前列腺增生(对照组)组织中PTEN和COX-2的表达水平及其微血管密度(MVD).结果 PCa组PTEN阳性表达率明显低于对照组(P<0.05),而COX-2阳性表达率明显高于对照组(P<0.05);并且随着Gleason评分和临床分期的提高,PTEN的表达明显下降、COX-2的表达明显升高(均P<0.05).PCa组MVD值明显高于对照组(P<0.05);在PCa组中,高Gleason评分患者MVD值明显高于低Gleason评分患者(P<0.05),但在不同临床分期组差异无统计学意义(P >0.05).PTEN阴性者和COX-2阳性者MVD值均明显高于PTEN阳性者和COX-2阴性者(P<0.05);PTEN和COX-2间呈明显负相关(P<0.05).结论 PTEN基因的缺失和COX-2的高表达是PCa发生、发展的重要原因,且与肿瘤的Gleason评分和临床分期密切相关;它们之间可能存在协同作用,共同促进肿瘤血管生成.

Abstract:Objective To investigate the expression of PTEN and COX-2 in prostate cancer and their relationship with angiogenesis. Methods The expressions of PTEN,COX-2 and CD34 in 60 PCa samples were analysed by tissue microarray technology and immunohistochemistry,and compared with those of 25 BPH samples. Results The expressions of PTEN were observed in 33.3%,50.0% and 100% of PCa,HPIN and BPH respectively (P〈0.05),and those of COX-2 were 66.7%,42.9% and 4.0% respectively (P〈0.05).The expression of COX-2 increased with the increases of Gleason degree and clinical staging (P 〈0.05), while more loss of PTEN with the increases of Gleason degree and clinical staging (P〈0.05).The expression of PTEN was negatively related to COX-2 in PCa.Of the PCa samples,MVD counting in PTEN negative group or COX-2 positive group was higher than that in PTEN positive group or COX-2 negative group (P〈0.05). Conclusion PTEN and COX-2 proteins are significantly correlated with clinicopathotogical characteristics and biologic behaviours in PCa,and they may play a role in angiogenesis.
