糖基化终末产物对人脐静脉内皮细胞凋亡及其受体表达的影响

摘    要:目的 研究糖基化终末产物(AGEs)对人脐静脉内皮细胞(HUVEC)凋亡及其受体(RAGE)表达的影响。方法 采用流式细胞仪技术,荧光显微镜,电镜观察AGEs对培养的HUVEC凋亡的影响,同时用RT-PCR方法检测RAGE的mRNA的表达。结果 AGEs加入细胞培养中,可明显诱导HUVEC凋亡,并呈剂量依赖效应关系。在发生凋亡的同时有RAGE mRNA表达的增强。结论 AGEs对HUVEC有诱导凋亡的作用,该作用可能通过RAGE介导。

Abstract:Objective To explore the mechanism of advanced glycosylation end products (AGEs) in damaging human umbilical venous endothelial cells (HUVEC) and affecting the expression of receptor of AGEs (RAGE). Methods The effect of AGEs on apoptosis of cultured HUVEC was studied by flow cytometry technique, fluoresence microscope and electron microscope. The expression of RAGE mRNA was also assayed by RT-PCR. Results The ratio of apoptosis was 6.5% in control, but increased to 12.8%, 31.2%, 93.4% when treated with 10, 50, 100 mg/L of AGEs. The effects were dose-dependent. RAGE mRNA expression was much higher in apoptosis cells than that in normal cells. Conclusion AGEs induce apoptosis in HUVEC, which might be mediated by RAGE.
