胃癌病人外周血单个核细胞替代癌细胞的体外化疗药敏试验

目的　用胃癌病人外周血单个核细胞 (PBMCs)替代手术切除标本进行化疗药物敏感性测定 ,以辅助解决胃癌病人个体化疗用药问题。②方法　取胃癌病人手术标本的同时抽取其外周血 ,采用噻唑蓝 (MTT)法 ,体外检测 2 7例胃癌病人癌细胞和PBMCs对 7种化疗药物的敏感性。③结果　不同化疗药物对胃癌病人癌细胞、PBMCs抑制率差异均有显著性 (F =5 .2 8,5 .17,P <0 .0 1) ;但每一种化疗药物对胃癌病人癌细胞和PBMCs抑制率差异均无显著性 (t =0 .92 44～ 1.6 6 98,P >0 .0 5 ) ;胃癌病人癌细胞抑制率与PBMCs抑制率间呈正相关关系(r=0 .6 978～ 0 .886 4,P <0 .0 1)。④结论　用PBMCs替代癌细胞进行化疗药物敏感性测定 ,可为胃癌病人个体化疗用药提供一条新的途径。

A STUDY OF SENSITIVITY TEST OF CHEMOTHERAPEUTIC AGENTS BY USING PBMCs ALTERNATIVE CANCER CELLS IN GASTRIC CANCER PATIENTS

Objective To study the sensitivity of chemotherapeutic agents by using PBMCs alternative cancer cells in patients with gastric cancer and screen the sensitive agents for individual chemotherapy. Methods The chemosensitivity of PBMCs and cancer cells in 27 gastric cancer patients to seven chemotherapeutic agents were detected by using a tetrazolium based colorimetric assay (MTT assay). Results The inhibitory effects of chemotherapeutic agents to cancer cells or PBMCs of gastric cancer patients were obviously different( F=5.28, 5.17, P <0.01), but the average inhibitory effects between cancer cells and PBMCs to each chemotherapeutic agent were not obviously different ( t =0.924 4-1.669 8, P >0.05). The average inhibitory effects between cancer cells and PBMCs to the same chemotherapeutic agent showed positive correlation( r =0.697 8-0.886 4, P <0.01). Conclusion Using PBMCs alternative cancer cells to detect chemosensitivity of chemotherapeutic agents will provide a new way for screening the sensitive agents for individual chemotherapy.
