| 摘 要： | 目的探讨睫状神经营养因子CNTF在正常大鼠和猫脊髓中的表达.方法从正常成年SD大鼠及成熟猫脊髓组织中抽提总RNA,管家基因NF-M为内参,RT-PCR技术检测CNTF mRNA脊髓组织中的表达.结果(1)用RT-PCR技术检测到了CNTF mRNA在正常大鼠脊髓中的表达;(2)用相同的RT-PCR条件未能检测到CNTF mRNA在正常猫脊髓中表达.结论(1)在正常大鼠脊髓中有CNTF mRNA表达,提示CNTF可能与脊髓的生理功能有关;(2)CNTF mRNA可能在猫脊髓中不表达;用于扩增大鼠CNTF的RT-PCR条件可能不适合于猫;或大鼠和猫的CNTF mRNA存在种属差异.

| Abstract: | Objective To explore the expression of the CNTF mRNA in spinal cord of normal adult rats and cats.Methods The spinal cord of normal adult srague-daweley(SD) rats and cats at the lumber segments were taken out respectively.Total RNA was extracted with TRIzol reagent,and CNTF gene was amplified by RT-PCR technique.Results(1)CNTF mRNA is observed in spinal cord of normal SD rats by RT-PCR;(2) With the same RT-PCR as rats,CNTF mRNA cannot be detected expression in spinal cord of normal adult cats.Conclusion(1) The expression of CNTF mRNA was detected in spinal cord of normal adult SD rats,which indicated that CNTF might be involved in the physiological process of neurons in spinal cord;(2)CNTF mRNA might not express in spinal cord of normal adult cats at all;the amplification reaction performed in rats was not suit to cats;or there were genera diversity of CNTF mRNA between rats and cats. |
