鲨鱼软骨制剂抗肿瘤作用的研究

摘    要:以鲨鱼软骨为原料,经盐酸胍抽提,丙酮分级沉淀,超滤,Sephadex G-75柱层折等步骤得到鲨鱼软骨制剂 1(SP1)和鲨鱼软骨制剂 2(SP2)。测定它们对肿瘤细胞和血管内皮细胞DNA合成的抑制效应,对小鼠移植瘤的抑癌作用,以及对小鼠脾脏和胸腺的影响。结果表明:SP1能同时抑制血管内皮细胞和直接抑制肿瘤细胞;SP2同时具有提高机体免疫功能、直控抑制肿瘤细胞和抑制血管内皮细胞的活性。整体实验说明它们都具有较强的抑制肿瘤生长的活性,对小鼠体重无明显影响,无明显毒副作用,显示了良好的应用前景。

Abstract:Shark-cartilage preparation 1 (SP1) and 2 (SP2) were isolated from shark cartilage by the method of guanidine hydrochloride extraction, acetone fractioned precipitation, ultrafiltration and chro-matography. SP1 inhibited the DNA syntheses of endothelial cell and Hela cell, and didn't inhibit that of QGY7703 cell and human skin fibroblast. SP2 inhibited the DNA syntheses of endothelial cell, Hela cell and QGY7703 cell, but didn't inhibit that of human skin fibroblast, when the concentration was 20 tag/ml. SP2 inhibited all that of the 4 cell lines at the concentration of 60 pg/ml. SP1 and SP2 also inhibited the growth of tumors implanted on mice, and SP2 stimulated their spleen and thymus in vivo, but both of them have no effect on mice weights. These results suggest that SP1 and SP2 have antitumor effect, and may be an active drug to carcinoma therapy in some day.
