四物汤对3种血虚证动物细胞周期的影响

摘    要:目的系统考察60Coγ-射线、环磷酰胺和综合放血法致血虚证小鼠模型骨髓细胞周期变化的特点。方法采用60Coγ-射线照射、环磷酰胺腹腔注射和综合放血法制作血虚证动物模型,检测对小鼠骨髓细胞周期的影响,同时观察四物汤给药后对其影响。结果射线损伤所致血虚证小鼠骨髓细胞中G2/M期比例先升高后降低,环磷酰胺损伤所致血虚证小鼠骨髓细胞中G2/M和S期细胞先升高后降低;综合放血组小鼠骨髓细胞的S期细胞数先升高后降低;四物汤可以促进射线损伤致血虚证小鼠骨髓细胞G0/1期进入S期的作用。3种模型都先后出现了G2/M期阻滞,以γ-射线照射组出现最早,与照射组不同的是环磷酰胺组与综合放血组在造模后都先后在第3、7天出现了S期细胞峰值。结论3种模型骨髓细胞周期都出现了G2/M期阻滞,但S期的变化存在明显不同,四物汤可以促进射线损伤致血虚证小鼠骨髓细胞G0/1期进入S期的作用。

Abstract:Objective To investigate the changes of cell cycle of bone marrow in blood-deficient mice induced by 60Coγ irradiation,cyclophosphamide and compound bleeding. Methods Blood deficiency models were established by using 60Coγ irradiation,cyclophosphamide and compound bleeding.Changes of cell cycle was detected by PI labeling, and the effects of Siwutang on cell cycle were also detected. Results Cell cycle of bone marrow cells in BD induced by irradiation in G2/M phase increased at first and then decreased, so did cell cycle of BD induced by cyclophosphamide in G2/M and S phase and cell cycle of BD induced by compound bleeding in S phase. Siwutang could promote cell cycle from G0/1 to S phase, but had little effect on cell cycle of BD induced by cyclophosphamide and compound bleeding. Conclusion Cell cycle of three BD animal models arrested G2/M.The change of S stage was of different nature in the pathogenesis and development, and Siwutang could promote cell cycle from G0/1 to S phase.
