头穴电针对脑出血大鼠脑组织GLUT-1及共基因表达的影响

摘    要:目的:研究大鼠脑出血后血肿周围组织葡萄糖载体蛋白-1(GLUT-1)、GLUT-1mRNA表达的动态变化规律及头穴透刺治疗作用。方法:将雄性Wistar大鼠144只随机分为假手术组、模型组、针刺组,每组又分为24h、72h、168h三个小组,采用胶原酶/肝素联合注入法制备脑出血大鼠模型。针刺组给予百会透太阳穴电针治疗,按指定时间断头取脑,取血肿周围3mm范围内组织,分别采用免疫组织化学法检测GLUT-1和RT-PCR法检测GLUT-1mRNA表达的变化。结栗:脑出血后,模型组三个时间点均见大部分GLUT-1阳性微血管形态变窄、皱缩、闭塞,越靠近病灶越严重;与假手术组比较,模型组GLUT-1及GLUT-1mRNA表达24h时失代偿性升高(P〈0.05),72h、168h时失代偿性降低(P〈0.05)。针刺组:仅在24h时血肿周围出现了轻微的管腔狭窄,之后大部分血管表现为管腔呈圆形扩张:GLUT-1与GLUT-1mRNA表现相同的变化趋势,即与假手术组比较,24h时变化不明显,72h、168h时出现降低(P〈0.05),且各时间点与模型组有明显差异(P〈0.05)。结论:头穴电针能扩张血管,改善供血,减轻血管内皮细胞的损伤,故能推迟GLUT-1及GLUT-1mRNA病变的发生,延长治疗时间窗。

Effects of Scalp Electroacupuncture on Cerebral GLUT-1 and GLUT-1 mRNA Expressions in Intracerebral Hemorrhage Rats

Abstract:Objective: To study the dynamic changes of GLUT-1 and GLUT-1 mRNA expressions of cerebral tissues in intracerehral hemorrhage rats and the acupuncture's effects on them. Methods: 144 Wistar rats were divided into 3 groups: sham group, model group, and acupuncture group. Every group was subdivided into three groups in time (24h, 72h, 168h). The rat model of intracerehral hemorrhage was made with Collagense VII/heparini infusion, and treated by electro-acupuncture at acu- points Baihui (GV20) through Taiyang (EX-HN1). Immunohistochemical method was used to observe the expressions of GLUT-1 and RT-PCR method to detect GLUT-1 mRNA expression on the perihemotoma. Results: After intracerehral hemorrhage, model group showed narrowing, crimpling and obliteration in majority of GLUT-1-positive micro-vessels in all three times, more severe in nearer loci. In comparison with sham group, the expressions of GLUT-1 and GLUT-1 mRNA compensatorily rose within 24h(P 〈0.05), and fell in 72h and in 168h (P〈0.05). In acupuncture group: a few GLUT-1-positive micro-vessels were a bit narrowed in 24h, and they were normal in other times; compared with sham group, the expressions of GLUT-1 and GLUT-1 mRNA changed little in 24h time, and obviously fell in 72h and 168h times, but there were significant differences in comparison with the model group (P〈 0.05). Conclusion: Scalp electroacupuncture can dilate the vessels, improve blood supply, lessen impairment to endothelial cells, and hence delay GLUT-1 and GLUT-1 mRNA expressions to prolong treatment time window.
