补体抑制免疫复合物沉淀作用的实验研究

本文以辣根过氧化物酶（ＰＯ）作抗原，与相应抗体（抗－ＰＯ）混合，加入血清，对血清补体抑制免疫复合物沉淀作用（ＩＩＰＣ）进行了研究。结果，在抗原抗体最适比例时形成免疫复合物最多；原血清及１：２稀释血清ＩＩＰＣ最强，ＯＤ值为０．５７和０．５４，血清的进一步稀释对ＩＩＰＣ有一定影响；孵育时间以３０ｍｉｎ最宜，ＯＤ值为０．５６；离心时间在２５～４５ｍｉｎ时ＯＤ值最大为０．５０～０．５６；在最适条件下，正常人混合血清ＯＤ值为０．４９；５６℃３０ｍｉｎ灭活血清为０．１４；５０℃１５ｍｉｎ处理血清为０．３８；ＥＤＴＡ处理血清为０．０９；ＥＧＴＡ处理血清为０．２１；对２０例健康人血清ＩＩＰＣ测定结果为０．３９～０．６２，平均０．４８±０．０７。表明正常血清补体对免疫复合物的沉淀有明显抑制作用。各种因素处理血清对ＩＩＰＣ均有一定影响，而以５６℃３０ｍｉｎ和ＥＤＴＡ处理血清最明显。文中还对该法的影响因素进行了讨论。

EXPERIMENTAL STUDY ON INHIBITION OF IMMUNE PKECIPITATION BY COMPLEMENT

In this paper,a method for measuring inhibition of immune precipitation bycomplement (IIPC) using peroxidase(PO)as an antigen was studies. The results showed thatimmune complex of PO and anti-PO rabbit IgG at equivalence could form a great quantity ofprecipitation. OD values were 0. 57 and 0. 54 in non and diluted serum (1 : 2),0. 56 in 30 minincubation time,and 0.50～0.56 in 25～45 min centrifugation time. OD value of normal indi-viduals mixture serum ,the serum of heating to 56C for 30 min ,50℃ 15min ,and treated by-ethylenediamine tetracetate (EDTA) ,ethyleneglycol tetracetate(EGTA ) were 0.49,0.14,0, 38,0. 09,0. 21 respectively. IIPC of 20 normal persons was 0. 48 + 0. 07(0. 39～0.62).These results indicate that normal serum markedly inhibites precipitation of immune complexand many agents may block IIPC of serum ,especially 56℃ for 30 min and EDTA,The vari-ous factors of inhibiting IIPC are discussed in the paper.
