循环miRNA21在肝细胞癌早期诊断中的价值

目的 探讨血浆中miRNA21作为肝细胞癌早期生物标志物的诊断价值.方法采用定量RT-PCR（quantitative RT-PCR）检测55例肝细胞癌患者、60例慢性肝炎患者、50例健康对照者血浆中miRNA21的表达水平.结果 血浆miRNA21在肝细胞癌组中水平显著高于慢性乙肝组（P〈0.01）和健康对照组（P〈0.01）.通过绘制受试者工作曲线（receiver-operator characteristic curve,ROC curve）分析,miRNA21作为肝细胞癌和慢性乙肝的诊断标志物,其诊断价值分别为：灵敏度（65%）,特异度（85%）,受试者工作曲线下面积（AUC）为0.771,而miRNA21作为肝细胞癌和健康对照者的诊断标志物,其诊断价值分别为：灵敏度（89%）,特异度（95%）,AUC为0.956.结论 miRNA21是肝细胞癌潜在的早期诊断标志物.

Circulating microRNA-21 as a novel biomarker for hepatocellular carcinoma

Objective The aim of this study was to evaluate the significance of plasma microRNA-21 level as a biochemi- cal marker for hepatocellular carcinoma. Methods Plasma microRNA-21 level was measured by RT-PCR. And it was compared in three groups： 55 patients with hepatocellular carcinoma, 60 patients with chronic hepatitis, and 50 healthy volunteers. Results Plasma microRNA-21 level in 55 patients with hepatoeellular carcinoma was significantly higher than in patients with chronic hepatitis and healthy volunteers （P 〈 0.01, P 〈 0.01, respectively）. Receiver-operator characteristic curve analysis of plasma mi- cro-RNA-21 yielded an AUC of 0.771 with 65% sensitivity and 85% specificity when differentiating hepatoeellular carcinoma from chronic hepatitis, and an AUC of 0. 956 with 89% sensitivity and 95% specificity when differentiating hepatocellular carcinoma from healthy volunteers. Conclusion Plasma microRNA-21 level might be a promising biochemical marker for hepatocellular car- cinoma.
