氢气对氧化低密度脂蛋白诱导的内皮细胞凋亡的抑制作用

摘    要:目的探讨氢气(H2)饱和培养液对氧化低密度脂蛋白(ox-LDL)诱导的内皮细胞凋亡的影响。方法人脐静脉内皮细胞株EA-hy926分为:ox-LDL组,ox-LDL+不同浓度H2组和正常对照组;流式细胞仪检测细胞凋亡率和细胞内ROS水平;Western blot检测NF-κB p65蛋白水平。结果 ox-LDL+H2组(氢浓度0.6 mmol/L)与ox-LDL组相比,细胞凋亡率明显降低(2.48±0.21)%vs(4.01±0.39)%,P<0.01],细胞内ROS含量减少(2.05±0.22)vs(5.32±1.04),P<0.01],NF-κB p65蛋白水平降低(0.29±0.02)vs(0.66±0.07),P<0.01],且上述作用随H2浓度增加而增强。结论 H2饱和培养液可减少ox-LDL诱导的内皮细胞凋亡,其机制可能与减少ROS含量和抑制NF-κB激活有关。

Abstract:Objective To study the effect of hydrogen(H2)-rich culture fluid on apoptosis in endothelial cells induced by oxidized low-density lipoprotein(ox-LDL).Methods Endothelial cells from human umbilical veins were divided into control group,ox-LDL group,and H2 group(ox-LDL + H2-rich fluid).Apoptosis of endothelial cells and level of intracellular reactive oxygen species(ROS) were assayed by flow cytometry.NF-κB p65 protein was detected by Western blotting.Results The apoptosis rate of endothelial cells,intracellular ROS level,and NF-κB p65 protein level were significantly lower in ox-LDL group than in 0.6 mmol/L H2 group(2.48%±0.21% vs 4.01%±0.39%,2.05±0.22 vs 5.32±1.04,0.29±0.02 vs 0.66±0.07,P<0.01),which increased with the increasing H2 concentration in culture fluid.Conclusion H2-rich fluid can protect endothelial cells against ox-LDL-induced apoptosis by reducing intracellular ROS level and inhibiting NF-κB activation.
