前列腺良、恶性病变组织中细胞增殖与细胞凋亡表达的相关性研究

摘    要:目的 :研究前列腺良、恶性病变组织中细胞增殖与细胞凋亡的关系及其生物学意义。方法 :对 36例前列腺癌(Pca)、2 0例前列腺增生 (BPH)和 10例正常前列腺 (NP)的细胞增殖和凋亡情况进行研究。应用免疫组织化学ABC法检测增殖细胞核抗原 (PCNA)来反映细胞增殖活性 (PI) ,用原位末端标记 (TUNEL)的方法反映细胞凋亡 (AI)情况。结果 :PI和AI在前列腺癌组织中的表达明显高于良性前列腺增生 (P <0 0 1) ,PCNA与前列腺癌分级有关 ,随着肿瘤分级增高而增高 ;前列腺增生组织中细胞增殖活性较正常前列腺明显增高 ,但细胞的凋亡率却显著下降。结论 :细胞增殖与细胞凋亡的增加在前列腺癌的发生和发展中起重要作用 ;而前列腺组织细胞增殖的增加和细胞凋亡的减少参与了BPH形成过程。

Abstract:Objective:To investigate the relationship between the rate of proliferation and apoptosis index to further understand the tumor biology of prostate cancer.Methods:The proliferation status was evaluated by using monoclonal antibodies which recognize the proliferation cell neclear antigen(PCNA).The apoptosis status was detected by the terminal deoxynucleotidyl transterase-mediated dUTP biotin nick end-labeling(TUNEL).The total of 36 cases prostate cancer,20 cases benign prostatic hyperplasia and 11 cases of normal prostate specimens were studied.Results:The proliferation index and apoptosis index of prostate cancer were much higher than these of benign prostatic hyperplasia;The proliferation index of BPH was much higher than that of normal prostate,and the apoptosis index was much lower than that of normal prostate.Conclusion:The results suggest that both high proliferation rate and apoptosis rate are associated with the pathogenesis of prostate cancer;Both high proliferation rate and low apoptosis rate are associated with the pathogenesis of BPH.
