摘    要:目的研究表达重组人白细胞介素15(rhIL-15)工程菌高密度发酵的条件。方法采用发酵罐发酵,对影响工程菌生长和目的蛋白质表达的因素进行优化。结果以甘油为碳源可明显抑制有害物质乙酸的积累,提高工程菌的表达量和收获率。高密度发酵明显提高了目的蛋白质的表达量和工程菌的产量。结论该发酵工艺大大提高了rhIL-15工程菌的产量和表达水平,为rhIL-15的规模化生产打下了基础。

Abstract:Purpose To investigate the optimal high cell density fermentation condition of recombinant human interluekin-15 in E. coli. Methods The effects of the pH range, air input, incubation time and fedbatch carbon sources on the expression level of rhIL-15 and cell output were analysed. Results With the glycerol as the carbon sources it inhibited the accumulation of acetic acid and increased the yield of E. coli and the expression level of rhIL-15. High cell density fermentation enhanced the output of E. coli and the expression level of rhIL-15 significantly. Conclusion The optimized fermentation condition for recombinant E. coli raised significantly the high expression level of rhIL-15. It is suitable for the fermentation of rhIL-15 on a large scale.
