甲基化在肺癌中的意义

摘    要:目的探讨基因T-钙黏蛋白(CDH13)在40例癌组织,40例的癌旁组织标本和非肺癌良性肺切除组织标本15例作为对照的表达的临床意义,进一步研究其基因异常甲基化与肺癌的相关性。方法采用特异性甲基化PCR(MSP)检测该基因在肺癌组织中异常甲基化的水平。结果 1.在肺癌组织中CDH13基因甲基化率明显高于癌旁组织和非肺癌良性肺切除组织,甲基化率分别为32.5%、7.5%、6.7%,有统计学意义(P<0.05)。2.CDH13基因异常甲基化和CDH13的表达水平呈负相关性(r=-0.520).3.CDH13基因启动子区5’CpG岛甲基化阳性率在肺癌组织中显著高于癌旁组织和非肺癌良性肺切除组织,CDH13基因启动子区5’CpG岛甲基化导致了CDH13的表达下调。结论在肺癌组织中CDH13基因异常甲基化率明显升高且有临床诊断意义,CDH13基因在癌组织中的异常甲基化水平将有望成为肺癌新的治疗靶点。

Abstract:Objective The aim of this study was to investigate the clinical significance of trullcated-cadherin(CDH13) expression in 40 cases of tumour sample,40 cases of cancer-surrounding tissues and 15 specimens of normal tissues,further to research potential relationship between abnormal gene methylation and lung cancer.Methods It was used to detect CDH13 by RT-PCR.The detecting of the5′methylation of CDH13 gene promoter CpG islands was quoted methylation specificity PCR method,observing the result used spectrophotomete after agarose gel electrophoresis.Results 1.There was significant different in the group of lung carcinomas tissue,cancer-surrounding tissues and normal lung tissues.The rate of CDH13 gene methylation respectively is 32.5%,7.5%,6.7% with a statistical significance(P〈0.05).2.There was significantly negative correlation among the methylation of CDH13 gene and the expression of CDH13(r=-0.520) 3.The positive rate of the5′methylation of CDH13 gene promoter CpG islands obviously is higher in the lung cancer organization than in the group of cancer-surrounding tissues and normal lung tissues.There was obviously related.in the5′methylation of CDH13 gene promoter CpG islands and the reduces positive rate of CDH13 gene expression.Conclusion It is clinical significance of the expression level of CDH13 to diagnose lung carcinoma,and the expression level of CDH13 in ung carcinomas may be a novel promising strategy for the treatment of lung carcinoma.
