大鼠坐骨神经切断后丙戊酸钠对相应脊髓运动神经元p-ERK1/2表达的影响

摘    要:目的 研究坐骨神经切断后丙戊酸钠(VPA)对相应脊髓运动神经元p-ERK1/2表达的影响.方法 选取健康成年雄性Wister大鼠64只,随机分为对照组和实验组,分别于术后1、3、7、14d四个时相处死后取患侧L4-L6脊髓前角,应用免疫组织化学、Western Blot技术检测p-ERK1/2在脊髓前角运动神经元中的表达变化.将两组所得数值进行统计学分析.结果 Western Blot实验结果 显示对照组:术后3d时,p-ERK1/2的表达达到高峰;术后7-14d,p-ERK1/2的表达逐渐下调;实验组:术后3d时,p-ERK1/2的表达达到高峰;术后7-14d,p-ERK1/2的表达逐渐下调;术后1-7d,实验组相关p-ERK1/2表达明显高于对照组(P<0.05).结论 VPA对脊髓运动神经元的保护作用可能是因为VPA激活了MAPK/ERK信号传导途径.

Abstract:Objective To investigate the Effect of valproic acid on expression of phosphor-ERK1/2 in motor neurons in spinal cord after sciatic nerve injury in rats.Methods Adult male wister rats were randomly divided into the control group and the experimental group.In the experimental group,the right sciatic nerve was transected and survived for 1,3,7 and 14d after operation.The changes in the expression of p-ERK1/2 in L4-5 spinal cord were detected by immunohistochemistry and western blot methods.Results In the control group,p-ERK1/2 expression has increased and reached the peak in 3 days after sciatic nerve transection.The p-ERK1/2 expression began to decrease at 3 days.In the experimental group,p-ERK1/2 expression has increased and reached the peak in 3 days after sciatic nerve transection.The p-ERK1/2 expression began to decrease at 3 days.The p-ERK1/2 expression in the experimental group is higher than that in the control group in 7 days.Conclusion Protective effect of valproic acid on motor neurons in spinal cord is likely by activating MAPK/ERK1/2 signaling pathway.
