3味清热中药调节兔动脉粥样硬化相关因素的实验研究

摘    要:目的 观察具有清热作用的葛根、黄连、决明子对兔动脉粥样硬化(AS)相关指标的调节作用.方法 将家兔按血脂水平随机分为空白组、模型组、阿托伐他汀组、葛根组、黄连组及决明子组.空白组动物给予普通饲料,其余各组动物饲喂高脂饲料.高脂饲料喂养期间各组动物按体质量分别给予相应剂量药物.给药12周后,测定血脂水平、血清抗氧化能力、凝血及血液流变学指标,并观察动脉斑块形成程度.结果 葛根、黄连可显著降低AS兔主动脉弓部斑块校正面积(PA/CVA)与最大斑块厚度(MPT),降低AS兔血清总胆固醇(CHO)、低密度脂蛋白胆固醇(LDL-C)及非高密度脂蛋白胆固醇(non-HDL-C)水平;决明子则可降低血清三酰甘油(TG)水平.葛根与决明子可降低AS兔血清丙二醛(MDA)水平,升高超氧化物歧化酶(SOD)活力.葛根、黄连与决明子均可降低AS兔血小板最大聚集率;葛根与黄连可降低低切变率全血黏度.结论 具有清热作用的葛根、黄连、决明子有较好的抑制AS形成的作用,与其降血脂、升高机体抗氧化能力、抑制血小板聚集、改善血液流变学等作用有关.

Abstract:Objective To observe the effect of Radix Puerariae Lobatae, Rhizoma Coptidis and Semen Cassiae, which possessing clearing heat function, in regulating atherosclerosis indexes of rabbits. Methods The rabbits were randomly divided into six groups: blank control group, model control group, atorvastatin group, Radix Puerariae Lobatae group, Rhizoma Coptidis group and Semen Cassiae group, according to the serum lipid level. The rabbits in blank control group took ordinary feeds and others took high fat feeds. The rabbits were given corresponding dose of the drug for twelve weeks. Subsequently to measure the serum lipid level, serum antioxidant ability, blood coagulation, hemorheology and observe the pathological arteriosclerosis plaque lesions. Results Radix Puerariae Lobatae and Rhizoma Coptidis reduced the plaque area corrected by cross-sectiona vessel wall area (PA/CVA) and the max plaque thickness (MPT) at the aortic arch in arteriosclerosis rabbits. Radix Puerariae Lobatae and Rhizoma Coptidis reduced the serum CHO, LDL-C and non-HDL level in arteriosclerosis rabbits, while Semen Cassiae reduced the serum TG level. Radix Puerariae Lobatae and Semen Cassiae not only reduced the serum MDA level but increased the SOD activity. All of the three drugs decreased the max platelet aggregation rate, and Radix Puerariae Lobatae and Rhizoma Coptidis reduced whole blood viscosity of low shear rate. Conclusion The traditional Chinese medicines which possessing clearing heat function can inhibit the atherosclerosis formation by reducing blood lipids, promoting anti-oxidant ability, inhibitting platelet aggregation and improving hemorrheology.
