生米卡链霉菌DNA对北里链霉菌原生质体种间转化

摘    要:采用SDS-苯酚法提取生米卡链霉菌DNA,同时以SDS处理生米卡链霉菌原生质体得到游离DNA。将提取DNA及游离DNA转化北里链霉菌原生质体,得到10~(-2)的转化率。转化子经发酵得柱晶白霉素,其产量比亲本高5.2倍,同时发现组份亦有所变化,A_5组份从9.1%提高到36.7%。

Abstract:This paper reports the abstracted DNA and dissociated DNA of S. mycarofaciens transformed the protoplasts of S. kitastoensis.It was a success to abstract the DNA of S. mycarofaciens by means of SDS-phe-nol and meantime to get the dissociated DNA by treating the protoplasts of S. kitas-atoensis with SDS. The protoplasts of 5. kitasatoensis were transformed by abstracted DNA and dissociated DNA and got 10-2 transformation rate. The products of the transformants were analysed by bioanalysis and HPLC and the results suggested that they were leucomycin and the output were higher than that of parent. The highest one was 5.2 times than that of parent. Meanwhile, there were some bigger component's changes in which component A5 increased from 9.1% to 36.7%,These consequences indicated that transformation could increase the output and improve the component of antibiotic.
