X射线诱导Raji细胞凋亡的实验研究

摘    要:目的探讨X射线对人Burkkit淋巴瘤细胞株Raji细胞增殖及凋亡的影响。方法用不同剂量X射线照射细胞,并在不同时间段用倒置显微镜观察照射后细胞的形态变化及生长抑制率,流式细胞仪检测细胞凋亡率、细胞周期,并观察X射线对克隆形成的影响。结果 16Gy照射后48h对Raji细胞的抑制率(92.67±1.20)%最显著;8Gy照射后48h早期凋亡率(42.04±3.62)%最高;8Gy照射后24h细胞周期G2/M期细胞(57.86±3.31)%,明显大于正常对照组(14.54±2.32)%;8Gy照射后第7天无克隆形成,第14天克隆数为(5.33±2.40),明显小于正常对照组第7天(116.67±20.28)和第14天(263.33±20.27)。结论 X射线可抑制细胞增殖,并诱导细胞凋亡,照射后细胞周期阻滞在G2/M期,X射线8Gy照射后不能完全抑制Raji细胞的增殖。

Abstract:Objective To investigate the effects of X-ray irradiation on the proliferation and apoptosis of human Burkkit lymphoma cell line Raji.Methods The cells were irradiated with various doses of irradiation.After irradiation,the morphology of the irradiated cells was observed under microscope.Cell cycle and apoptosis were assessed by flow cytometry.Results At 48 hours after irradiation(16 Gy),the percentage of growth inhibition was(92.67±1.20)%.The proportion of early apoptotic cells induced by 8 Gy irradiation at 48 h was greatest(42.04±3.62)%.At 24 hours after 8 Gy irradiation,the proportion of cells(57.86±3.31)% arrested at G2/M-phase was significantly greater than the control group(14.54±2.32)%.Results from the clonogenicity assay showed that the number of colony after 8 Gy irradiation(5.33±2.40) was lower than the control group at day-7(116.67±20.28) and day-14(263.33±20.27).Conclusion X-ray irradiation can inhibit the proliferation,induce cell cycle arrest,and induce apoptosis of Raji cells.X-ray irradiation may also inhibit the growth of cancer stem cells.
