无锡市2009年副溶血性弧菌脉冲场凝胶电泳分型研究

摘    要:目的分析无锡市2009年副溶血性弧菌分子特征,以及细菌性食物中毒分离的菌株与菌株之间、食源性疾病监测分离的副溶血性弧菌之间的遗传相关性,建立无锡地区副溶血性弧菌分子分型数据库,为无锡市进行食源性疾病的快速反应和预警、监测和控制提供科学依据。方法脉冲场凝胶电泳(PFGE)对菌株进行分子分型,所得结果以BioNumericsV4.0软件UPGMA方法进行聚类分析。结果 21株菌以SfiⅠ酶切后可分为9个PFGE型别,各群间相似性系数〉60%。其中有两起食物中毒菌株DNA指纹图谱完全相同,食源性疾病监测分离株分属7个型别。3型和6型为全年的优势型别。结论 3型和6型为无锡市2009年副溶血性弧菌优势型别,需加强防范。

Molecular typing of Vibrio parahaemolyticus by pulsed field gel electrophoresis in Wuxi city in 2009

Abstract:Objective To study the characteristics of genotypes of Vibrio parahemolyticus strains collected in Wuxi city in 2009,analysis the phylogenic relationship among the strains isolated from the samples of bacterial food poisoning and foodborne illness, and establish the molecular typing databases of Vibrio parahemolyticus, so as to provide scientific evidence for quick reaction and early warning, monitoring and control of foodborne illness. Methods Pulsed field gel electrophoresis(PFGE) was used for molecular typing of isolates. Clustering of patterns was analyzed with software BioNumerics. Results All 21 strains were clustered into 9 PFGE patterns when digested with Sfi I,with similarity coefficient between groups more than 60%.The strains of DNA fingerprinting of two food poisoning incidents were identical,the strains isolated from foodborne illness belonged to 7 PFGE patterns.Subtype 3 and subtype 6 were predominant patterns in 2009. Conclusions The predominant patterns were subtype 3 and subtype 6 in Wuxi in 2009. It is necessary to enhance the prevention of bacterial food poisoning and foodbourne illness.
