bcl—2基因在良性前列腺增生中的作用

摘    要:利用前列腺组织特异性的基因载体新型C3(1)和转基因技术,培植出人bcl2基因的前列腺组织特异性转基因鼠。免疫组织化学检测证实人bcl2蛋白在鼠前列腺上皮细胞中的过量表达。病理学观察发现,转基因鼠已发生良性前列腺增生。该实验为说明bcl2基因在良性前列腺增生(BPH)中的病理作用提供了最直接的证据,并繁殖出可稳定传代的BPH的动物模型。

Abstract:To establish an animal model for prostate hyperplasia,three lines of transgenic mice were generated by using the prostate specific rat C3(1) promotor to drive expression of the human bcl 2 cDNA. The tissue specific overexpression of human bcl 2 protein in prostatic epithilium was confirmed by immunohistochemical analysis.Male transgenic mice develop atypical benign prostate hyperplasia by 10 months of age in up to 50% of animals. The experiment provides the immediate evidence of the critical role of protooncogene bcl 2 in the development of BPH.Moreover, these transgenic mice offered an suitable animal system to study the etiology of BPH.
