| 摘 要： | 目的研究PTEN基因表达对乳腺癌细胞生长增殖的影响。方法将分别携带有野生型、突变型PTEN基因的真核表达载体pBP—wt—PTEN以及pBP—G129R—PTEN，以脂质体介导法转人人PTEN表达缺失的乳腺癌细胞ZR-75—1中，嘌呤霉素筛选获得稳定表达PTEN基因的转染细胞，通过细胞活力实验、流式细胞术观察PTEN基因对ZR-75—1细胞生长增殖的影响。结果稳定转染PTEN基因的ZR-75—1细胞中不仅PTEN蛋白稳定表达，而且其生长速度较对照组明显减慢。流式细胞术显示，细胞周期发生G1期阻滞。结论外源性PTEN基因导入ZR-75—1细胞后，可引起ZR-75—1细胞生长阻滞，抑制肿瘤细胞增殖。
