空肠弯曲菌在免疫小鼠体内粘附和定居的初步研究

摘    要:本文用空弯菌CJ-S_(131)抗血清和甲醛处理的菌苗分别免疫小鼠,活菌攻击后,以体内CFU法研究该菌在免疫小鼠体内的粘附和定居。用特异性抗血清、IgG之Fab段、热敏抗原抗血清(ALAS)被动免疫小鼠后,该菌在小鼠肠内的粘附和定居均受到显著的抑制;而耐热抗原的抗血清(ASAS)则无显著抑制效果。主动免疫小鼠抗该菌粘附和定居的能力和肠粘膜免疫功能强弱有关。一次免疫组肠固有膜内特异性抗体形成细胞(AFC)较加强免疫组少,故加强免疫组抗空弯菌CJ-S_(131)粘附和定居的能力较强,CFU/g显著低于对照组。本文结果表明,CFU/g能较客观地反映粘膜免疫状态,因此,体内CFU法可作为估价粘膜免疫功能的一种研究方法。

Abstract:Mice were passively and actively immunized with the antiserum against C. jejuni, strain CJ-S131, and the oral vaccine of formalin-treated CJ-S131, and then were challenged with the live bac-teia to study on its intestinal adherence and colonization of the mice by the colonyforming units(CFU)method in vivo. Bacterial ad-herence was significantly inhibited by immunizing the mice with specific amtiserum, Fab-fragments of the IgG and anti-heat-labile antigen serum(ALAS) but not anti-heat-stable antigen serum(ASAS). Ability to inhibitintestinal adherence and colonization of active immunized nice paralleled to the ammounts of antibody-forming cells(AFC)in the lamina propria, more of which could be detec-ted in the secondarily primed mice and less in the primarily primed mice. Spe-cific Fab-fragments could inhibit bacterial autoaggregation in vitro of CJ-S131. The results showed that adhesion of CJ-S131 was heat-labile and could induced mu-cosal immunity preventing CJ-S131 from adhesion and colonization. It was sug-gested that CFU per gram intestinal tissue could truly reflect the state of mucosal immunity of gut, thus, the CFU method in vivo might be used as one methods to evalute the functionally mucosal immunity.
