摘    要:目的 介绍急性神经元贴壁的简易方法和应用激光扫描共聚焦显微镜观察药物作用后神经元内核酸代谢的方法。方法 以双重荧光染料吖啶橙 (Acridineorange ,AO)进行染色。在 36°C孵育箱内孵育 15分钟。激光扫描共聚焦显微镜对染色后的细胞进行光束扫描 ,以观测细胞内核酸代谢变化的规律。结果 皮层神经元RNA有显著改变。结论 本方法的建立可以成功地应用于激光扫描共聚焦显微镜对胞内核酸代谢变化的研究。

Abstract:Objective To introduce the simple method for acute-separated neuron adhibitting and the method to investigate the alteration of nuclear acid metabolism after drugs action with Laser scanning confocal microscope(LSCM).Method The cell was dyed with fluorescent dying AO,and scanned with LSCM to observe the alteration rule of nuclear acid.Results The RNA has significant alteration in morphine depended animals than that in control group.Conclusion The establishment of this method can be successfully used to study the alteration of nuclear acid with Laser scanning confocal microscope.
