姜黄素对NOD糖尿病小鼠Th1细胞和CD4^+Treg细胞的作用

摘    要:目的研究姜黄素对Th1细胞和CD4^+Treg细胞的作用,探讨姜黄素对非肥胖糖尿病(NOD)小鼠的治疗作用。方法将10周龄雌性NOD小鼠分为对照组和姜黄素组,分别用溶剂或姜黄素灌胃。第14周时测血糖。第20周时,流式细胞术分析外周血Th1细胞和CD4^+Treg细胞,分析胸腺CD4^+ CD8^-T细胞、CD8+CD4^-T细胞及CD4^+Treg细胞;分选脾CD4^+Treg细胞用于检测其抑制功能。结果姜黄素组血糖正常比例高于对照组。姜黄素组Th1细胞比例显著低于对照组(P<0.05),CD4^+Treg细胞比例与对照组差异无统计学意义(P>0.05),CD4^+Treg/Th1显著高于对照组(P<0.05)。姜黄素组胸腺CD4^+CD8^-T细胞、CD8^+CD4^-T细胞及CD4^+Treg细胞比例与对照组差异无统计学意义(P>0.05),胸腺总细胞数显著高于对照组(P<0.05)。姜黄素组CD4^+Treg细胞的抑制功能明显高于对照组(P<0.05)。结论姜黄素降低NOD小鼠外周Th1细胞、增加CD4^+Treg细胞抑制功能、缓解胸腺退化,延缓了NOD小鼠的高血糖症状。

Effects of curcumin on Th1 cells and CD4~+ Treg cells in non-obese diabetic female mice

Abstract:Objective To study the effect of curcumin on Th1 cells and CD4^+Treg cells,and explore the therapeutic effect of curcumin on type 1 diabetes.Methods 10 week-old female non-obese diabetic(NOD)mice were divided into control group and curcumin group,which were administered with solvent or curcumin by gavage.At 14 week-old,the blood glucose was measured.At 20 week-old,Th1 cells and CD4^+Treg cells of peripheral blood were analyzed by flow cytometry.Meanwhile,CD4^+CD8^-T cells,CD4-CD8+T cells and CD4^+CD8^-CD25^+T cells in thymus were analyzed also by flow cytometry.Then,spleen CD4^+Treg cells were sorted to evaluate the suppressive function in vitro.Results The proportion of mice with normal blood glucose was higher in curcumin group than that in control group.The proportion of Th1 cells in peripheral blood was lower in curcumin group than that in control group,whereas there was no difference in CD4^+Treg cells of peripheral blood between two groups.But the ratio of CD4^+Treg cells/Th1 cells increased in curcumin group.Although there were no significant differences in the proportions of CD4^+CD8^-T cells,CD8^+CD4^-T cells and CD4^+Treg cells between two groups,the number of total cells in thymus was higher in curcumin group than that in control group.Importantly,the suppressive function of CD4^+Treg cells increased significantly in curcumin group.Conclusions Curcumin reduces peripheral Th1 cells,increases the suppressive function of CD4^+Treg cells,and reduces thymus degeneration,which might play roles in delaying hyperglycemia symptoms in NOD mice.
