基于H-NMR技术的甲状腺乳头状癌血清代谢组学研究

摘    要:目的探讨不同生物学特征甲状腺乳头状癌(PTC)患者及微小乳头状癌是否存在阶段化代谢组学特征。方法对南京医科大学第二附属医院普外科2018年1月至2018年6月期间,甲状腺微小乳头状癌(PTMC)合并颈部淋巴结转移患者(T1组)、PTMC未合并颈淋巴结转移患者(T2组)、PTC合并颈部淋巴结转移者(T3组)、PTC未合并颈部淋巴结转移患部者(T4组)、健康对照者(HC组)血清进行基于H NMR技术的代谢组学分析。结果经O PLS DA分析表明,四组甲状腺乳头状癌患者血清与健康对照组血清比较,其代谢物图谱均能得到很好的区分,分别鉴定出9种、6种、10种和7种差异代谢物。 在这些代谢产物中,以氨基酸代谢途径中代谢物为主。结论不同生物学特征的甲状腺乳头状癌患者及微小乳头状癌存在阶段化代谢组学特征,提示这些代谢物可作为潜在血清代谢标记物。

The serum metabolic profiling on different biological behavior of patients with papillary thyroid carcinoma
Abstract:Objective To explore the staged metabolic characteristics of different biological characteristics of patients with papillary thyroid carcinoma. Methods Metabolomics analysis was performed in serum from PTMC patients with positive cervical lymph (T1 group), PTMC patients with negative cervical lymph (T2 group), papillary thyroid carcinoma (PTC) patients with positive cervical lymph (T3 group), PTC patients with negative cervical lymph (T4 group), and healthy control (HC group). Results Subsequent analysis demonstrated exclusively altered metabolic profiles in these diseases groups: a total of 9, 6, 10 and 7 differential metabolites were identified to distinguish from HC group, respectively. Among these metabolites, amino acid metabolism emerged as the most significantly disturbed pathway. Conclusion Those metabolites including Lactic acid, glutamic acid, lysine and alanine may serve as potential biomarkers to differentiate PTMC from PTC and HC.
