hTERT在人胰腺癌组织中的表达及其与COX-2、P-gp、Bcl-2蛋白表达和临床病理特征的关系

摘    要:目的研究端粒酶逆转录酶(hTERT)、环氧合酶-2(COX-2)、P-糖蛋白(P-gp)和Bcl-2蛋白在胰腺癌组织中的表达及其相互关系,以及hTERT与胰腺癌的病理特征的关系。方法选择胰腺癌38例,慢性胰腺炎25例,正常胰腺组织20例。以SABC法检测hTERT、COX-2、P-gp和Bcl-2的表达。按手术记录描述肿瘤的病理特征。结果在38例胰腺癌组织中hTERT、P-gp、COX-2、Bcl-2阳性表达率分别为81.58%、52.63%、76.32%、57.90%,评分分别为(6.04±1.08)分、(4.50±1.75)分、(5.96±1.60)分、(5.61±1.08)分,而正常胰腺组织和慢性胰腺炎组织的hTERT与Bcl-2阳性表达率和评分均为0,在慢性胰腺炎P-gp、COX-2阳性率分别为20.00%、24.00%,评分分别为(1.21±0.86)分、(3.59±1.42)分,在正常胰腺组织阳性率分别为15.00%、0,评分分别为(0.90±0.58)分、0分。胰腺癌组与其他组各项阳性表达率和评分比较差异均有统计学意义(P〈0.05)。在胰腺癌组织中,hTERT表达与P-gp(rp=0.365,P〈0.05)、COX-2(rp=0.534,P〈0.001)、Bcl-2(rp=0.557,P〈0.001)表达和临床分期(rp=0.333,P〈0.05)呈显著正相关。结论hTERT、P-gp、COX-2和Bcl-2蛋白在胰腺癌组织中均有较高表达,hTERT表达与P-gp、COX-2、Bcl-2表达和胰腺癌的临床分期均有显著的正相关性。

Abstract:Objective To investigate the expression of hTERT protein and be correlation with the expressions of COX-2,P-gp,Bcl-2 protein by immunohistochemistry in human pancreatic cancer (PC) tissues,and be correlation with pathological features of PC.Methods Thirty-eight cases of pancreatic cancer,25 cases of chronic pancreatitis and 20 cases of normal pancreatic tissues were studied.Expressions of hTERT,P-gp,COX-2 and Bcl-2 proteins were determined by SABC of immunohistochemistry.The pathological features of PC were decribed by according to operation records.Results The positive rate of expressions of hTERT,P-gp,COX-2,Bcl-2 protein were 81.58,52.63,76.32,57.90 respectively,and their scores were 6.04±1.08,4.50±1.75,5.96±1.60,5.61±1.08 respectively in 38 pancreatic cancer tissues,but expressions of hTERT and Bcl-2 protein were none in chronic pancreatitis tissues and normal pancreatic tissues (P〈0.001).The positive rates of expression of P-gp and COX-2 were 20.00 and 24.00 and their scores were 1.24±0.86 and 3.59±1.42 in chronic pancreatitis tissues respectively.The positive rates of expression of P-gp and COX-2 were 15.00 and 0 and their scores were 0.98±0.58 and 0 in normal pancreatic tissues respectively (P〈0.05).There were positive correlations between expression of hTERT and P-gp(rp =0.365,P〈0.05) or COX-2 (rp=0.534,P〈0.001)or Bcl-2 (rp =0.557,P〈0.001).Expression of hTERT had to do with clinical stages (rp =0.333,P〈0.05),but not to do with site,size and histological classification (P〉0.05).Conclusions There were higher expressions of hTERT,P-gp,COX-2 and Bcl-2 in pancreatic cancer tissues than in others.There were positive correlations between expression of hTERT and expressions of P-gp,COX-2,Bcl-2 protern and the clinical stage of PC.
