摘    要:目的建立同时测定走马胎中3种三萜皂苷类成分的RP-HPLC-ELSD方法。方法选用HPLC Kromasil C18色谱柱(4.6mm×250mm,5μm),流动相:甲醇55%CH3OH90%CH3OH梯度洗脱40 min,流速0.8 mL·min-1。Althche 2000ES ELSD检测器,漂移管温度:100。C,空气流速2.8 L·min-1。结果结果表明3-O-{-L-吡喃鼠李糖基-(1→3)--D-吡喃木糖基-(1→2)]--D-吡喃葡萄糖基-(1→4)--D-吡喃葡萄糖基-(1→2)]--L-吡喃阿拉伯糖基}-西克拉敏A(a),3-O-{-L-吡喃鼠李糖基-(1→3)--D-吡喃木糖基-(1→2)]--D-吡喃葡萄糖基-(1→4)--D-6-O-乙酰基吡喃葡萄糖基-(1→2)]--L-吡喃阿拉伯糖基}-西克拉敏A(b),3-O-{-L-吡喃鼠李糖基-(1→3)--D-吡喃葡萄糖基-(1→3)--D-吡喃木糖基-(1→2)]--D-吡喃葡萄糖基-(1→4)--D-吡喃葡萄糖基-(1→2)]--L-吡喃阿拉伯糖基}-16-羟基-13,28-环氧-齐墩果烷(c)的检测分别在3.00090%CH3OH梯度洗脱40 min,流速0.8 mL·min-1。Althche 2000ES ELSD检测器,漂移管温度:100。C,空气流速2.8 L·min-1。结果结果表明3-O-{-L-吡喃鼠李糖基-(1→3)--D-吡喃木糖基-(1→2)]--D-吡喃葡萄糖基-(1→4)--D-吡喃葡萄糖基-(1→2)]--L-吡喃阿拉伯糖基}-西克拉敏A(a),3-O-{-L-吡喃鼠李糖基-(1→3)--D-吡喃木糖基-(1→2)]--D-吡喃葡萄糖基-(1→4)--D-6-O-乙酰基吡喃葡萄糖基-(1→2)]--L-吡喃阿拉伯糖基}-西克拉敏A(b),3-O-{-L-吡喃鼠李糖基-(1→3)--D-吡喃葡萄糖基-(1→3)--D-吡喃木糖基-(1→2)]--D-吡喃葡萄糖基-(1→4)--D-吡喃葡萄糖基-(1→2)]--L-吡喃阿拉伯糖基}-16-羟基-13,28-环氧-齐墩果烷(c)的检测分别在3.0009.000μg,8.6859.000μg,8.68537.635μg,3.30037.635μg,3.30010.450μg线性关系良好。药材中三种成分的平均回收率(n=9)为99.64%(RSD 0.78%),99.93%(RSD 0.65%),100.07%(RSD 1.37%)。结论该方法操做简单、结果准确具有较好的重复性和稳定性,为走马胎药材质量控制提供了一个很好的依据。

Abstract:Objective To establish the quantitative method of three triterpenoid saponins in Ardisia gigantifolia Stapf by HPLC-ELSD. Methods The column was packed with 5 pm Kxomasil C18 stationary phase. The mobile phase consisted of methanol-water, eluted in gradient mode of 55% CH3OH-90%CH3OH, flow rate was 0.8 mL.min-1.The temperature of drift tube was 100~Gand the air flow rate was 2.8 Lomin1. Results The linear ranges of the three components were 3.000-9.000 μg, 8.685-37.635pg and 3.300-10.450pg ,respectively. The average recoveries of the three components in Ardisia gigantifolia Stapfwere 99.64 %(RSD=0.78 %), 99.93 % (RSD=0.65%), 100.07 %(RSD=1.37%) ,respectively. Conclusion It was proved that the method was reliable, simple, and precise, that could be used for quality control.
