热水提取罗非鱼鱼皮胶原蛋白的研究

摘    要:目的研究热水水解法提取罗非鱼鱼皮胶原蛋白的最佳工艺及检测鱼皮胶原蛋白的氨基酸组成。方法采用热水水解提取法,探讨于不同温度、pH值、提取时间和液固化等因素对罗非鱼鱼皮胶原蛋白提取的影响,并和多因素正交试验确定最佳工艺流程。结果罗非鱼鳞胶原蛋白热水提取最佳提取工艺为温度90℃,pH值6.0,固液比1:13,时间2h,蛋白得率为212mg/g原料。特征氨基酸羟脯氨酸为9.7%。结论研究建立的热水水解法提取罗非鱼皮胶原蛋白纯度高、色泽洁白无异味。

The extraction of tilapia skin collagen by using hot water

Abstract:Objective To explore the methods for r extracting collagen from tilapia skin and the components of amino acid of collagen. Methods The tilapia skin collagen was extracted by using hot water. Results The optimum conditions were the extraction temperature of 90℃, the pH value of 6.0, liquid/solid ratio of 1:13 and extraction time of 2h. The highest extraction rate of collagen protein from Tilapia skin was 265.3mg per gram raw material. The rate of oxyproline hydroxyproline (the typical amino acid of collagen protein) reached 9.7%. Conclusion The mothod is easy and feasible.
