VEGF抑制血清撤离诱导的颊癌细胞凋亡的初步研究

摘    要:目的 体外实验中初步观察VEGF对血清撤离诱导的人颊鳞癌BCaCD885细胞凋亡的影响。方法 分别将处于对数生长期的BCaCD885细胞培养于完全RMP11640(10%胎牛血清)、无血清RMPI1640、含VEGF(50ng/ml)的无血清RMPI1640,48h后观察不同培养条件下癌细胞形态变化及用流式细胞仪检测细胞凋亡发生比率。结果 无血清RMPI1640培养组BCaCD885细胞凋亡增加,而含VEGF(50ng/ml)的无血清RMPI1640培养组的细胞凋亡率低,两之间差异有显性(P<0.01)。结论 外源加入的VEGF可抑制血清撤离诱导的颊癌细胞的凋亡,提示VEGF在口腔恶性肿瘤的发生发展中起着多重的促进作用。

Abstract:Objective The suppression effect of VEGF on the apoptosis induced by serum deprivation in oral squamous cell carcinoma was observed. Methods BCaCD885 buccal mucosa squamous cell carcinoma cells were cultivated for 48h under serum-deprived culture medium (0% FBS), serum-deprived culture medium (0% FBS) with rhVEGF165 (50ng/ml) or normal culture medium (10% FBS), and observed the change of cell morphology and the rate of cell apoptosis detected by FCM. Results Forty-eight hours after culture with serum-deprived culture medium, more BCaCD885 cells were undergo apoptosis than those cultured in the normal culture medium. But the rate of apoptosis of BCaCD885 cells dropped to 0.19% after culture 48h under serum-deprived culture medium with rhVEGF165. Conclusion VEGF prevents the apoptosis of the BCaCD885 cells cultured in the serum-deprived medium. The result indicated that VEGF not just as an angiogenesis factor, but also as an anti-apoptosis factor during the development of oral malignant tumor.
