主动载药法制备槐定碱脂质体

摘    要:目的:考察槐定碱脂质体不同制备方法对其包封率的影响。方法:采用注入、薄膜分散、逆向蒸发等被动载药法和硫酸铵梯度、pH梯度结合逆向等主动载药法制备槐定碱脂质体,用高速冷冻离心-HPLC法测定脂质体包封率。结果:被动载药法制备的脂质体包封率较低,主动载药法中pH梯度结合逆向法制备脂质体包封率为93.27%。结论:采用主动载药法可制得包封率较高的槐定碱脂质体。

Abstract:Objective: To study the influence of different preparation methods on the entrapment efficiency. Methods: To prepare sophoridine liposomes by passive loading method and active loading method, the entrapment efficiencies were determined by high speed freeze centrifugation-HPLC method. Results: The entrapment efficiency with passive loading method was low, while up to 93.27% with active loading method. Conclusion: The sophoridine liposomes with high entrapping efficiency can be prepared by pH-gradients method coupled with reverse evaporation technique.
