桑叶总黄酮对大鼠心肌缺血再灌注损伤的保护作用

摘    要:目的:研究桑叶总黄酮对大鼠心肌缺血再灌注损伤心肌细胞凋亡及半胱天冬酶-3(caspase-3)蛋白表达的影响。方法:90只SD大鼠随机分为假手术组、模型组、桑叶总黄酮(TFMF)高、中、低3个剂量组,通心络胶囊组。手术前1周,MSTF服用不同剂量的MSTF(35,70,140 mg.kg-1.d-1),通心络胶囊组按照100 mg.kg-1.d-1服用,其余2组给与等量的生理盐水。采用结扎左冠状动脉前降支30 min、再灌注2 h的方法建立大鼠心肌缺血再灌注损伤模型。观察心肌梗死面积,原位末端标记(TUNEL)染色检测心肌细胞的凋亡情况,免疫组织化学法检测caspase-3蛋白表达情况。结果:与假手术组比较,模型组IS/LV,IS/AR增加,细胞凋亡率显著增加(P<0.01),且caspase-3蛋白表达显著升高(P<0.01);与模型组比较,不同剂量的TFMF能降低IS/LV,IS/AR,明显减少心肌细胞凋亡(P<0.05或P<0.01),明显降低caspase-3蛋白表达(P<0.05或P<0.01)。结论:TFMF能下调caspase-3蛋白表达,减少细胞凋亡而发挥抗心肌缺血再灌注损伤作用。

Abstract:Objective: To study effects of the total flavones from Mori Folium(TFMF)on myocardial cell apoptosis and caspase-3 expression in myocardial ischemic-reperfusion injury in rats. Method: Ninety SD rats were randomized into sham operation group,model group,three dose groups of TFMF(35, 70, 140 mg·kg-1·d-1),Tonxinluo capsule group(100 mg·kg-1·d-1).The left anterior descending coronary artery was ligated for 30 min, followed by 2 h of reperfusion to establish the rat model of myocardial ischemia-reperfusion injury. myocardia apoptosis were detected by the TUNEL staining and caspase-3 expression was detected by immunohistochemical technique method. Result: IS/LV and IS/AR increased obviously in model group compared with those in sham group, Myocardial apoptosis rate increased obviously(P<0.01)and caspase-3 protein expression was significantly elevated(P<0.05 or P<0.01)in the model group. Compared with model group,IS/LV,IS/AR and myocardial apoptosis rate in TFMF group decreased obviously(P<0.05 or P<0.01),and caspase-3 protein expression was down-regulated(P<0.05 or P<0.01). Conclusion: TFMF possessed an obviouse ameliorate effect on myocardial cell apoptosis in myocardial ischemic-reperfused injury rats by decreasing the caspase-3 protein expression.
