痰瘀同治方对大鼠颈总动脉粥样硬化易损斑块稳定性的影响

摘    要:目的:探讨痰瘀同治方对大鼠颈总动脉粥样硬化(AS)易损斑块稳定性的影响。方法:采用高脂、高蛋氨酸喂养,4周末行颈总动脉球囊拉伤术造模,术后将大鼠随机分为空白对照组、模型组、辛伐他汀组、复方维生素组、痰瘀同治方(TYTZ)7.8,3.9,1.95 g.kg-1组,连续ig 8周。实验结束后,测定各组血清同型半胱氨酸(Hcy)、血清基质金属蛋白酶-9(MMP-9),基质金属蛋白酶组织抑制因子-1(TIMP-1)和斑块中MMP-9,TIMP-1阳性表达。结果:与模型组比较,辛伐他汀组、复方维生素组和痰瘀同治方7.8,3.9 g.kg-1组血清MMP-9降低并TIMP-1升高,斑块中MMP-9阳性表达降低。结论:痰瘀同治方可通过调节MMP-9/TIMP-1起到稳定颈总动脉粥样硬化易损斑块的作用。

Abstract:Objective: To observe the effects of Tanyu Tongzhi(TYTZ) formula on stability of vulnerable atherosclerotic plaque of carotid artery in rats. Method: With high cholesterol lipid and 2% L-methionine feeding for four weeks, the vulnerable atherosclerotic plaque model was reproduced by operation on carotid artery endothelium for inducing sacculus pull injury. Rats were randomized into five groups: normol control group, model group, simvastatin group, vitamin complex group, TYTZ group (7.8, 3.9, 1.95 g·kg-1) and administrated orally for 8 weeks. At the end, the blood sample was collected to measure the concentration of Hcy, MMP-9 and TIMP-1, the protein contents of MMP-2 and TIMP-2 in plaque were detected by immunohistochemistry. Result: In contrast to model group, TYTZ (7.8, 3.9 g·kg-1), simvastatin and vitamin complex decreased the concentration of serum MMP-9 and increased the concentration of TIMP-1, the expression of MMP-9 detected in plaque was also lower. Conclusion: TYTZ formula could stablize the vulnerable atherosclerotic plaque of carotid artery through mediating MMP-9 and TIMP-1.
