摘    要:目的建立高效液相色谱(HPLC)法测定人血浆中苦参碱的浓度。方法以0.02mol/L磷酸二氢钾溶液(含0.4%三乙胺,以磷酸调节pH值至6.0±0.1)-乙腈(88:12)为流动相,尼扎替丁为内标,血浆样品经二氯甲烷萃取后采用HPLC法测定。结果苦参碱质量浓度的线性范围是0.102~20.4μg/mL,平均相对回收率为93.3%~108,6%,日内和日间精密度RSD均小于10%,苦参碱的最低检测限为25ng/mL。结论该方法快速、准确、灵敏,可用于苦参碱的药代动力学研究。

Abstract:Objective To establish a HPLC method for detecting the concentration of matrine in human plasm. Methods The mobile phase consisted of 0. 02 mol/L potassium dihydrogen phosphate solution(containing 0.4% triethylamine,adjusted to pH 6. 0 ±0. 1 with phosphoric acid)- acetonitrile (88:12)was used. Nizatidine was used as the internal standard. The plasm samples were pretreated by dichloromethane, then determined by HPLC. Results The range of linear was 0. 102-20. 4 μg/mL. Both the intra- day and inter-day RSD were less than 10%. The recovery rate was 93.3% -108.6%. The minimum detective limit was 25 ng/mL. Conclusion The HPLC method is accurate,stable and sensitive. It can be applied to clinical pharmacokinetics study.
