生物素酶标法检测乙肝e系统的研究

摘    要:生物素、亲合素和 ELISA 结合,成为较普通法敏感、特异的检测法。实验中,生物素最佳标记用量为150μg/mg 抗-HBe,IgG 包被抗体、标记抗体最适应用浓度为10μg/ml。本法和普通ELISA 进行比较,检测 HBeAg、抗-HBe 的阳性率均较普通法高,检测 HBeAg 时,本法阳性、普通法阴性85份;检测抗-HBe 时,本法阳性、普通法阴性149份,经关联及相差分析,p<0.01。本法的特异性较好,检测 e 系统时批内、批间 CV 平均<10%。

Abstract:We used biotin-avidin system ELISA for the detection of hepatitis Be antigen and antib- ody.The results showed that this method was more sensitive and specific than the conventio- nal ELISA.During our experiment,BNHS 150 ug/mg Ab were used to label anti-HBe and the result was satisfactory.The optimum concentr- ation of both the coating and albelling antibo- dies were 10ug/ml.We compared BAS-ELISA with conventional ELISA and found that both HBeAg and anti-HBe positive rates were higher than the other.When HBeAg were measured, 85 sera were shown positive by BAS method but negative by conventional one.The specifi- city of BAS method was good.The CV of different groups and the CV of the same groups were less than 10%.
