应用BA—ELISA法定量测定IL—2

在 ELISA 基础上结合生物素—亲和素系统建立的 BA-ELISA 法用于定量测定 IL-2。该法以 McAb 作捕捉抗体,生物素化多抗为第2抗体,亲和素化酶起放大与呈色作用。其最小检测浓度2ng/ml,可检测线性范围256～2ng/ml。对其它小分子肽及蛋白制剂等无交叉反应,证明具有较强的特异性。同时对4种组合形式的 ELISA 法进行了 IL-2定量测定的比较,并简要探讨了此方法的影响因素。结果表明 BA-ELISA 法特异、快速,不需特殊条件与设备,适用于 IL-2的检测。

Quantitive Examination of IL-2 by BAELISA

BA-ELISA method was set up on the basis of combination of ELISA and biotin-avidin sys- tem and was used to detect IL-2 in quantity.In this method,McAb is capturing antibody,bioti- nyl polyclonal antibody is the second abtibody, enzyme labeled avidin plays a role in amplific- ation and coloration.The minimum concentrat- ion of detection is 2ng/ml and the linear detec- table range is 256-2ng/ml.There are no cross reactions with other small molecular peptide and protein,so the specificity is high.Four co- mbination forms of ELISA were campared for their quantity detection of IL-2,and the influ- nce factors were also discussed briefly.The re- sults showed that BA-ELISA method is rapid, highly specific and does not need special cond- ition and equipment.It is suited to the detect- ion of IL-2.
