摘    要:目的和方法:以离体心脏灌流法制备大鼠心肌缺氧、复氧模型,提取大鼠心矶mRNA并标记心肌线粒体肌酸激酶肌膜型亚基基因探针,进行Northern杂交,观察不同缺氧、复氧时sMiMiCKmRNA的变化及磷酸肌物保护作用。结果:随着缺氧时间的延长,sMiMi-CK的mRNA逐渐减少,复氧后sMiMi-CK进一步减少,PCr能显著改善sMiMi-CK的表达量。结论:PCr对缺氧、复氧的心肌具有明显的保护作用

Abstract:AIM and METHODS: Rat myocardial mRNA was extracted and mitochondrial sarcomeric creatine kinase (sMiMi-CK) was used as labled probed in vitro myocardial ischemia/reperfusion rats. Northern blot was performed to determine the changes of sMiMi-CK mRNA in different periods of ischemia/reperfusion and the protective effect of phosphocreatine (PCr).RESULTS:During early ischemial (10 min) mRNA was decreased about 18 24%. After reperfusion mRNA decreased again. PCr effectively prevented the decrease of mRNA expression of sMiMi-CK. CONCLUSION:The results indicated that PCr may possess protective effect on anoxic and reoxygenated myocardium.
