摘 要: | 目的观察选择性过氧化物酶体增殖物激活受体γ激动剂吡格列酮(Pioglitazone,PIO)对大鼠骨髓EPCs(Endothelial progenitor cells,EPCs)迁移的影响及并对其机制进行初步探讨。方法使用密度梯度离心法及差速贴壁法联合的方法培养大鼠骨髓EPCs,并使用双荧光染色的方法进行鉴定。将大鼠骨髓EPCs置于含不同质量浓度PIO的培养基中培养,检测EPCs的迁移情况。将培养7天的EPCs分5组,分别加入一定浓度的PIO;PPARγ拮抗剂GW9662;PIO及及PI3K/Akt通道阻滞剂Wortmannin;PIO及ERK通道阻滞剂PD98059;并设立空白对照组,检测EPCs的迁移情况。结果在PIO为50μmol/L时,可最大程度促进EPCs的迁移。这种作用在PI3K/Akt通道阻滞剂Wortmannin及PPAR-γ拮抗剂GW9662存在时作用消失,而在ERK通道阻滞剂PD98059作用下仍存在。结论 PIO可促进大鼠骨髓EPCs的迁移,这种作用可能是通过PI3K/Akt信号通路介导。

Abstract: | Objective TO explore selective peroxisome proliferator-activated receptor 7 agonists pioglitazone (PIO) to affect endothelial progenitor cells' (EPCs) migration and the mechanism. Methods EPCs were cultured using densi- ty gradient centrifugation and differential velocity adherent methods,and use double dyeing method to identify. Cultu- ring EPCs in medium containing pioglitazone of different concentrations, and setting up a blank control group, test EPCs' migration. EPCs are divided into 5 groups,respectively added to ploghtazone; ploglitazone and PPAR antago- nists GW9662; pioglitazone and PI3K/Akt channel blocker Wortmannin; pioglitazone and ERK channel blocker PD98059 ;and setting up a blank control group ,test EPCs' migration. Results When piogl.itazone at 50 b~mol/L con- centration,it can affect EPCs migration at the greatest degree. The effect disappeared when there are PI3K/Akt chan- nel blocker Wortmannin and PPAR-y? antagonist GW9662, but the effect didn,t disappeare when there is ERK channel blocker PD98059. Conclusion Pioglitazone can increase EPC migration. This effect may be mediated by PI3K/Akt sig nal pathway. |
