摘    要:目的探讨过氧化物酶体增殖物激活受体γ激动剂罗格列酮对内毒素(LPS)致大鼠肺组织炎症的影响及其作用机制。方法将36只SD大鼠随机分为生理盐水对照组(A组)、罗格列酮对照组(B组)、LPS模型组(C组),以及低、中、高剂量罗格列酮组(D组、E组、F组)。HE染色观察各组肺组织炎症;检测支气管肺泡灌洗液(BALF)中的细胞总数、中性粒细胞数、巨噬细胞数;酶联免疫吸附法检测BALF中的TNF-α、IL-1β和IL-8。结果与A组比较,C组肺组织炎症积分,BALF中的细胞总数、中性粒细胞数、巨噬细胞数及TNF-α、IL-8、IL-1β均明显增加;与C组比较,D、E、F组除IL-1β外,肺组织炎症积分,BALF中的细胞总数、中性粒细胞数、巨噬细胞数及TNF-α、IL-8均明显下降,E、F组与C、D组及F组与E组比较,均有统计学差异(P均〈0.05)。结论罗格列酮能下调TNF-α和IL-8表达,减轻肺组织炎症反应。

Influence of Rosiglitazone on pulmonary inflammation induced by endotoxin in rats

Abstract:Objective To investigate the influence and mechanism of Rosiglitazone ,an agonist of peroxisome proliferatots activated receptor gamma (PPAR-γ) ,on pulmonary inflammation induced by endotoxin in rats. Methods 36 male SD rats were randomly divided into 6 groups, namely saline control group( group A ), Rosiglitszone control group( group B) ,pulmonary inflammation group( group C), low dosage group of Rosiglitazone(group D), a moderate dosage group of Rosiglitazon (group E), high dosage group of Rosiglitazone( group F). The pulmonary inflammation scoring ,cell couts, the levels of tumor necrosis factor-or ( TNF-α), interleuldn-1β (IL-1β) and IL-8 in bronchoalveolar lavage fluid (BALF) were detected. Results Compared with group A, the pulmonary inflammation scoring, cell touts, the levels of TNF-α, IL-1β and IL-8 in BALF were significantly increased in group C ( P 〈 0.05). Compared with group C, these inflammation index except IL-1β were gradually decreased in group D, E, F. There were statistical difference of inflammation index among group D, E, F( P 〈 0.05 ). Conclusion Rosiglitazone can decrease the expressions of TNF-α, IL-8, and alleviate pulmonary inflammatiouin in rats.
