热毒平对热环境下小鼠腹腔巨噬细胞凋亡和Ca~(2+)浓度的影响

摘    要:目的探讨热毒平对巨噬细胞凋亡的影响及其机制,为中暑内毒素血症的治疗提供新的切入点和依据。方法在干球温度(34.5±0.5)℃、相对湿度(60±5)%的条件下建立小鼠中暑模型,将40只小鼠随机分为热毒平组、西黄芪胶组、生理盐水组、高温对照组和正常对照组,提取小鼠腹腔巨噬细胞,瑞士染色鉴定巨噬细胞,提取DNA行琼脂糖电泳,流式细胞仪检测凋亡指标和细胞内游离钙离子浓度。结果高温对照组、西黄芪胶组和生理盐水组小鼠腹腔巨噬细胞DNA电泳均出现明显的梯状条带,而热毒平组与正常对照组均未出现梯状条带。西黄芪胶组、生理盐水组和高温对照组小鼠腹腔巨噬细胞的凋亡率显著升高,热毒平组小鼠腹腔巨噬细胞的凋亡率明显低于这三组(P0.05),且与正常对照组接近(P0.05)。西黄芪胶组、生理盐水组和高温对照组小鼠腹腔巨噬细胞的Ca2+浓度明显升高(P0.05),热毒平组小鼠腹腔巨噬细胞的Ca2+浓度明显低于这三组(P0.05),且与正常对照组相近(P0.05)。结论热毒平能有效降低热环境下小鼠腹腔巨噬细胞的凋亡和巨噬细胞内Ca2+浓度,发挥抗中暑内毒素血症作用。

The influence of Reduping to the apoptosis and Ca^2+ concentration of macrophage under heat environment

Abstract:Objective To investigate the effect and mechanism of Reduping on macrophage apoptosis and provide experimental evidence and new method for treatment of endotoxemia of heat stroke. Methods Establishing the heat stroke model under circum stances of dry bulb temperature(34.5±0.5)℃ and(60±5)% relative humidity, 40 mice are randomly divide into five groups including Reduping group, Xihuangqijiao group, shengliyanshui group, heat stroke group and normal group. The apoptosis of groups using DNA fragmentation method and flow cytometry, Free Ca2 +concentration intracellular using flow cytometer were observed. Results The apoptosis of Reduping group is higher significantly(P0.05) than any other of three groups but similar to normal group; the free Ca^2+concentration was obviously less than any other of three groups but similar to normal group. Conclusion Reduping is reduce the apoptosis of macrophage and the level of free Ca^2+ concentration under heat environment effectively which can gainst endotoxemia of heat stroke.
