骨关节炎动物模型中99Tcm-枸橼酸的生物分布特征及其代谢研究

摘    要:目的利用骨关节炎动物模型观察 99Tcm-枸橼酸的生物分布和在骨关节炎症区域的代谢特点.方法 在制作骨关节炎动物模型的基础上,经在体显像及其感兴趣区技术计算器官和炎症关节的放射性活度的变化,同时也利用动物模型离体的测量器官或组织的百分注射量,分析 99Tcm-枸橼酸在各器官和骨关节炎症区域的代谢特点.结果 99Tcm-枸橼酸的蛋白结合率为(17.01± 0.75)%,骨关节炎症区域对 99Tcm-枸橼酸有较高的摄取,骨关节炎的患 /健比值出现在注射示踪剂后的 30 min到 1 h之间在体显像患 /健比值分别为 4.120± 0.552和 3.150± 0.587;百分注射量靶 /非靶比值分别为 3.279± 0.463和 3.899± 0.125.99Tcm-枸橼酸主要经泌尿系统排出,但也有少量是经肝脏-肠道系统排出.结论 99Tcm-枸橼酸有较低的蛋白结合率,是急性骨关节炎良好的显像剂,其最佳显像时间为注射 99Tcm-枸橼酸后的 1 h.

Abstract:AIM:To study the bio-distribution and metabolism in osteoarthritic area by using the osteoarthritic animal model. METHODS:On the basis of preparing the osteoarthritic animal model, the changes of radioactivity in the organs and inflammation joint area were determined by single photon emission computerized tomography(SPECT) and ROI technique.At the same time, the ID/g organ or ID/mL blood or urine was studied so as to analyze the metabolic characteristic of 99Tcm-citrate in the organs and bone and joint's inflammatory area. RESULTS:The combining rate of protein of 99Tcm-citrate was (17.01± 0.75)% .There was a high intake in the osteoarthritic area. The peak of lesion to no-lesion ratio appeared during 30 minutes to 1 hour after the injection of 99Tcm-citrate:the ratios of lesion to no-lesion determined by imaging were (4.120± 0.552) and (3.150± 0.587) respectively; the ratios of target to non-target of percentage injection volume were (3.279± 0.463) and (3.899± 0.125) respectively.99Tcm-citrate was mainly expelled from the urinary system, and also a little from liver-intestines system. CONCLUSION: The binding rate of plasma protein of 99Tcm-citrate is low. 99Tcm-citrate is a proper imaging agent for acute osteoarthritis, and the best time for imaging is 1 hour after its injection.
