| 摘 要： | 目的探讨MDR1反义寡脱氧核苷酸(Antisense oligodooxynucleotide,ASODN)片段逆转人胃腺癌耐药细胞SGC7901/ADM耐药的可行性。方法经MDR1 ASODN转染SGC7901/ADM细胞后,采用MTT法检测瘤细胞对阿霉素抗癌药的敏感性;采用流式细胞术检测细胞周期的改变、细胞内Rh123的潴留状况;采用免疫细胞化学检测MDR1蛋白质表达水平的变化。结果SGC7901/ADM细胞经MDR1 ASODN转染48小时后,对阿霉素化疗药物的敏感性增强,细胞对阿霉素的半数抑制浓度(IC50)由转染前的16.82μg/ml降至2.66μg/ml;与未经转染的SGC7901/ADM相比,ASODN/LF转染后出现明显的细胞周期阻滞,大量细胞被阻滞于S期;瘤细胞内RH123的潴留量显著高于转染前;瘤细胞内P-gp、MRP的表达显著下降。结论转染MDR1 ASODN可部分逆转人胃腺癌耐药细胞SGC7901/ADM耐药。
