糖尿病大鼠学习能力与脑细胞凋亡及血糖的关系

摘    要:目的探讨糖尿病大鼠学习能力与大脑神经元凋亡状况及血糖的关系。方法采用电迷宫法测试26只糖尿病大鼠(糖尿病组)和30只正常大鼠(对照组)的学习能力,并应用原位末端标记法检测其额叶皮层和海马发生细胞凋亡的神经元数目,免疫组化法检测凋亡调节基因Bcl 2、Bax的表达,并对糖尿病大鼠学习能力与额叶皮层和海马凋亡细胞数目及糖化血红蛋白进行相关分析。结果糖尿病组大鼠电迷宫测试成绩明显下降,达到学会标准前所需训练次数明显多于对照组(P<0.001),额叶皮层和海马细胞凋亡数高于对照组,Bcl 2表达减少,而Bax表达则增加(P均<0.01)。糖尿病大鼠达到学会标准前所需训练次数与额叶皮层和海马细胞凋亡指数呈正相关(r=0.410,P<0.05),且与糖化血红蛋白呈正相关。结论糖尿病高血糖状态下额叶皮层与海马凋亡调节基因表达改变引起的细胞凋亡增加可能与糖尿病认知功能障碍有关。
