胃癌组织中P-糖蛋白、肺耐药蛋白与化疗敏感性的相关性分析

目的:探讨P-糖蛋白(P-gp)、肺耐药蛋白(LRP)在胃癌组织中的表达及对体外培养胃癌原代细胞化疗药物敏感性的影响。方法:用逆转录-聚合酶链反应(RT-PCR)法检测胃癌组织中P-gp和LRP的编码基因mdr1和lrp的表达,用四氮唑蓝(MTT)快速比色法检测抗癌药物对胃癌原代细胞的IC50值。结果:与正常胃组织相比,mdr1和lrp在胃癌组织中的表达明显增加(P<0.01),两者之间mRNA表达量无相关性(P>0.05),但均与胃癌耐药密切相关(P<0.01)。结论:mdr1、lrp在胃癌组织中的高表达与肿瘤耐药密切相关,可作为肿瘤化疗时药物取舍的参考。

Relationship between the Expression of P-gp and LRP and Chemosensitivity in Gastric Cancer in Vitro

Objective To explore the expression of P - gp and lrp in gastric carcinoma, and their effect on chemosensitivity of cultured primary gastric cancer cells. Methods RT - PCR was performed to detect the expression of mdrl and lrp, and MTr assay was used to titer the IC50 of different antineoplasm drugs in cultured primary gastric cancer cells. Results Compared with control group,the mRNA expression of mdrl and lrp in gastric cancer,were increased significantly and there was no significant correlation between mdrl and lrp expression. However, mdrl, lrp expression were related with the drug resistance of gastric cancer. Conclusion The up expression of mdrl and lrp in gastric carcer were closely correlated with the drug resistance of gastric cancer, which could be as a reference to set an individual scheme of chemotherapy.
