补阳还五汤预先干预对大鼠脑死亡后心肺炎症细胞因子表达的抑制作用

摘    要:目的观察补阳还五汤预先干预大鼠抑制其脑死亡后心、肺炎症细胞因子表达。方法雄性Wistar大鼠30只,体质量180~200g,随机分3组。对照组:正常大鼠麻醉后取心和肺;脑死亡模型组:诱导大鼠脑死亡;补阳还五汤组:脑死亡诱导前7d,连续每天1次给大鼠ig补阳还五汤水煎剂(1.8mL/100g,12.6g/kg)。大鼠脑死亡诱导成功,机械呼吸6h,若平均动脉压大于80mmHg,则取其心和肺。RT-PCR检测心、肺组织肿瘤坏死因子-α(TNF-α)和白细胞介素-1β(IL-1β)mRNA表达,Western blotting检测心、肺组织TNF-α和IL-1β蛋白及磷酸化p38丝裂原活化蛋白激酶(p-p38MAPK)蛋白表达。结果模型组心肺TNF-α、IL-1βmRNA和蛋白表达及p-p38MAPK蛋白表达比对照组显著增加(P0.01)。补阳还五汤组各指标比模型组显著降低(P0.05),但仍比对照组显著增加(P0.01)。结论补阳还五汤预先干预大鼠能显著抑制其脑死亡后心肺炎症细胞因子表达,可能与其在不同层面阻断p38MAPK信号通路关键靶点有关。
